MALDI PMF is Matrix assisted laser laser desorption/ionisation Peptide mass fingerprinting, is an analytical protein identification technique. The peptide bonds are cleaved resulting in peptide smaller broken peptides whose masses are accurately measured. The theoretical peptide masses are measured by computerised protein sequences cut into peptides, and compared with the actual peptides. PMF algorithms assume peptides are from single source, thus, isolated proteins are taken for analysis. They are isolated in gel electrophoresis or SDS-PAGE combined with electroblotting or staining. A small fraction of peptide is added into the MALDI target and a chemical is added to the peptide mix for the desorption and ionisation of peptide molecules. This is initiated by pulsed laser beam, that transfers high among of energy into matrix molecules changing from solid phase to gas phase. The ions are accelerated towards electric field and fly towards detector, where they are detected as electric field.
