MALDI (Matrix assisted laser desorption/ionisation) is a Mass Spectrometry technique, for determination of proteins and peptides that make up the bio pharmaceuticals. The ionisation techniques gives us an accurate mass quantification, by high throughput quantification of biomolecules. This is done through ionisation by laser energy acts on the matrix of chemicals, that cuts the peptides and assimilates ions to from large molecules with minimal fragmentation. It allows fragile molecules to form bonds rendering intact also through covalent bonds that are protected from breaking. The matrix is a solid form creating ions by the laser light. The fragmentation is given by cleavage of peptide bonds that gives a specific peptide ion generated by set of fragments. This technique is used for molecular weight determination, monoclonal antibodies characterisation, analysis of covalent interactions through peptide drugs, stability analysis of chronically and physical bonds, peptide-protein conjugates of drugs. It is a fast technique less affected by contaminants, buffers and additives.
