LncRNA has about more than 200 nucleotides with low expression and stronger tissue specificity and do not encodes for proteins. In research, lncRNA regulates the coded genes expression with epigenetic inheritance, transcription and post-transcription for genetic improvement and diagnosis of occurrence and treatment. Different from traditional chip inspection, this method along with high-throughput sequencing and bioinformatics can sequence the LncRNA, this helps in quality assessment of RNA libraries, and coding peptides. Here, the total RNA detected by RNA degradation, contamination, detecting pure RNA for its concentration and integrity. Library construction with the poly-A of mRNA together with mRNA fragmentation to reduce rRNA reads after the total RNA is used for library. The cDNA is synthesized from fragmented RNA with reverse transcriptase and random primers and again converted to dsDNA with the buffer solution, dNTPs and polymerase. The cDNA fragments are repaired at the ends, ligated with adapters. This is used for Library detection and sequencing with initial quantification and dilution, and the fragment size of library is tested. LncRNA sequencing technology has been widely used in gene knock out over expression of lnc RNA genes, genetic improvement of species, and disease studies on occurrence, development, diagnosis, and treatment.
